About In Vivo Pharmacology
In vivo pharmacology studies test whether a compound does what it was designed to do in a living system: engaging its target, moving a disease-relevant endpoint, and doing so at exposures that can be translated to a clinical dose. They sit between in vitro screening and IND-enabling toxicology, and their results usually decide which candidate moves forward.
The work covers efficacy studies in disease models (oncology xenograft, syngeneic and PDX models, neurodegeneration, pain and behavioral models), dose-response studies, and target engagement or receptor occupancy measurements, often with PK/PD sampling built in. For antibodies and other biologics, the model species must express a target the molecule actually binds, which can narrow the choice of model and lab considerably.
Model validation matters more than the model name. Two labs can offer the same model with very different baseline variability, positive-control history and endpoint methods, so ask for recent control data before comparing on price.

- Can you share historical and positive-control data for this model from the past year?
- How are animals randomized, and are efficacy readouts scored blind to treatment?
- Can PK and biomarker samples be taken from the same animals to build a PK/PD relationship?
- How was group size chosen, and has a power calculation been done for the primary endpoint?
- For biologics: has target cross-reactivity been confirmed in this species or strain?
