In vitro work tests compounds in cells, tissues and enzyme systems rather than whole animals. It is fast, inexpensive and well suited to ranking many compounds, so it dominates early discovery and continues through development as part of the regulatory package.
The main areas are in vitro pharmacology (binding, functional and enzyme assays that measure potency and selectivity), in vitro ADME (metabolic stability, permeability, plasma protein binding and CYP inhibition and induction), and in vitro safety, where the hERG assay addresses cardiac risk under ICH S7B and the Ames and in vitro micronucleus assays form the first part of the ICH S2(R1) genotoxicity battery.
Ex vivo assays sit between in vitro and in vivo work: tissues or blood taken from dosed animals, or from human donors, are tested outside the body.
