About Oncology

Preclinical oncology CROs test whether a cancer therapy shrinks or slows tumors in animal models, at doses that are tolerated and exposures that can be reached in patients. Most work is done in mice, and the choice of model follows from the mechanism of the drug.

Cell-line xenografts grow human tumor lines in immunodeficient mice and are fast, reproducible and inexpensive. Patient-derived xenografts (PDX) keep more of the original tumor biology and are used to test response across many tumors. Syngeneic models implant mouse tumors into mice with intact immune systems, which immuno-oncology agents need, and humanized mice carry human immune cells for agents that act only on human targets. Orthotopic and metastatic models, often tracked with bioluminescence imaging, place tumors in the relevant organ.

Standard endpoints are tumor volume over time, tumor growth inhibition and survival, usually with PK and biomarker sampling to link exposure to effect. Combination studies with standard-of-care agents are common, so ask whether the CRO has dosed those agents before and holds historical control data for them.
