About Intrathecal Delivery

Intrathecal delivery places a test article directly into the cerebrospinal fluid of the subarachnoid space, bypassing the blood–brain barrier. It is central to preclinical programs for antisense oligonucleotides, AAV gene therapies and enzyme replacement therapies, where systemic dosing cannot reach the CNS at useful concentrations.

Studies range from single lumbar punctures to chronically implanted catheters with subcutaneous access ports for repeat dosing and CSF sampling. Nonhuman primates are the most common large-animal model because their spinal anatomy and CSF volume scale more closely to humans, but rodent, dog, pig and sheep models are also used. Our team has hands-on surgical experience with these procedures, and we use it to help sponsors judge which labs run them well.
