Preclinical immunology work falls into two groups. The first asks whether a drug harms or unintentionally activates the immune system. ICH S8 sets out immunotoxicity assessment, which typically starts with standard toxicology endpoints such as hematology, lymphoid organ weights and histopathology, and adds functional tests such as the T-cell dependent antibody response (TDAR), immunophenotyping by flow cytometry or NK cell activity when there is a signal or a mechanistic concern.
The second group asks whether a drug that targets the immune system works as intended. Immuno-oncology agents need models with a functioning immune system, such as syngeneic tumor models or humanized mice. Antibodies, bispecifics and engineered cell therapies need assays that measure immune cell activation, target engagement and cytokine release, often using human PBMC or whole blood in vitro alongside in vivo work. Autoimmune and inflammatory disease models, such as arthritis, colitis and experimental autoimmune encephalomyelitis, are offered by some labs; ask about them directly.
Flow cytometry capability is a good proxy for depth in this area: ask about panel size, validated species and whether the lab can analyze fresh samples on the day of collection.
