About Fc Effector Function Assays

Fc effector function assays measure what the constant region of an antibody does once it binds its target: antibody-dependent cellular cytotoxicity (ADCC) by NK cells, antibody-dependent cellular phagocytosis (ADCP) by macrophages, and complement-dependent cytotoxicity (CDC). Binding assays for Fc gamma receptors, C1q and FcRn complete the picture, with FcRn binding linked to half-life.

Whether effector function is wanted depends on the mechanism. Depleting antibodies need it; many blocking and agonist antibodies are engineered to remove it. Either way, regulators expect it to be characterized, and the assays are part of comparing lots and supporting biosimilars as well as discovery.

Assays can use primary effector cells, which are more physiological but vary between donors, or engineered reporter cell lines, which are more reproducible. Many programs use both.

Questions to ask when choosing a Fc Effector Function Assays CRO

  • Do you use primary effector cells, reporter assays, or both?
  • Which Fc gamma receptor variants and species orthologs can you test, including cynomolgus receptors?
  • How do you qualify the assays, and what reference antibodies run with each plate?
  • Can you run these assays to support lot release or biosimilarity comparisons?
